为了探索Rv0386做为转录因子时的功能，本文构建了Rv0386的重组表达载体并在大肠中表达得到了纯化后的蛋白。并由构建回补表达乳糖酶启动子的质粒，确定了Rv0386所在启动子区域，并由蛋白质和DNA结合实验确定Rv0386不能自调控。
