目的探讨卡介菌多糖-核酸对二硝基氟苯诱导的接触性皮炎的治疗作用,并探讨其可能的作用机制。方法利用二硝基氟苯诱导建立小鼠接触性皮炎动物模型。激发后48小时,肌肉注射卡介菌多糖-核酸。给药期间,隔日1次对激发部位进行评分,末次给药后24小时,取激发部位皮肤进行病理学观察,取外周血ELISA法检测血浆中IL-2,IL-4andIFN-γ水平,流式细胞术检测外周血中T淋巴细胞亚群的百分比。结果治疗作用研究结果表明:卡介菌多糖-核酸可剂量依赖性缓解二硝基氟苯诱导的小鼠接触性皮炎的症状,减少激发部位皮肤中炎症细胞的数目。作用机制研究结果表明:卡介菌多糖-核酸可剂量依赖性减少外周血中IL-4水平,增加IL-2和IFN-γ水平,增加外周血中CD4+T细胞百分比及CD4+T细胞与CD8+T细胞百分比的比值。结论卡介菌多糖-核酸有望成为一种有效的治疗接触性皮炎的药物,其作用机制可能与它调节T淋巴细胞亚群和细胞因子失调有关。
Objective To investigate the therapeutic effect of BCG-polysaccharide-nucleic acid on dinitrofluorobenzene-induced contact dermatitis and to explore its possible mechanism. Methods The animal model of contact dermatitis induced by dinitrofluorobenzene was established. Forty-eight hours after challenge, BCG polysaccharide-nucleic acid was injected intramuscularly. During the administration, the excitation site was scored once every other day, and the skin of the excitation site was observed for pathology 24 hours after the last administration. The level of IL-2, IL-4 and IFN- Cytometry to detect the percentage of T lymphocyte subsets in peripheral blood. Results Therapeutic effect The results show that: BCG-polysaccharide nucleic acid can dose-dependently alleviate the symptoms of contact dermatitis induced by dinitrofluorobenzene and reduce the number of inflammatory cells in the skin of the excited part. The results of the mechanism study showed that BCG-PSN could dose-dependently reduce the level of IL-4 in peripheral blood, increase the levels of IL-2 and IFN-γ, and increase the percentage of CD4 + T cells and CD4 + T cells in peripheral blood CD8 + T cell percentage ratio. Conclusion BCG polysaccharides - nucleic acid is expected to become an effective drug for treating contact dermatitis. Its mechanism may be related to its regulation of T lymphocyte subsets and cytokine imbalance.
