在制备电镜生物样品过程中,包埋剂的性能是一个十分重要的因素。选用不同的包埋剂浸透样品时,形成的包埋块的切割性能直接关系到超薄切片的成败。有些生物样品如藻类和植物的细胞具有较硬的细胞壁,有的组织则富含脂肪,还有某些样品需要用细胞化学的方法处理(如银染)。上述这类样品一般都不易浸透,影响着切片的质量。因此,选择适当的包埋剂是制做电镜样品中的一个必要条件。目前,国内常用的包埋剂主要是环氧树脂如国产618树脂,Epon812树脂,Spurr树脂和Ardldite 502树脂等。而我们实验室常用的是Epon812树脂混合剂和Spurr树脂混合剂这两种。Epon812树脂混合剂能较好地保存细胞的精细结构,有适当的反差,在电子束的轰击下相对稳定,并
In the preparation of electron microscopy biological samples, the performance of embedding agents is a very important factor. Selection of different embedding agent soaked samples, the formation of embedded block cutting performance is directly related to the success of ultra-thin slices. Some biological samples such as algae and plant cells have harder cell walls, some are fat-rich, and others require cytochemical methods such as silver staining. At present, the embedding agents commonly used in China are mainly epoxy resin such as domestic 618 resin, Epon812 resin, Spurr resin and Ardldite 502 resin. Epon812 resin mixture can better preserve the fine structure of cells, with appropriate contrast, relatively stable under the electron beam bombardment, and
