目的 The binding mechanism, morphological and conformational analysis on the nanocomposite of sea cucumber ovum derived octapeptide (EDLALLEK) with Ca2+well as its in vitro digestion and absorption behavior were investigated.方法 The interactions between the EDLAALEK and Ca2+were investigated by ITC, FTIR,RM and molecular simulation. The morphological analysis of the nanocomposite was investigated by using AFM, TEM and DLS. The corresponding conformational changes were studied by XRD and CD. Furthermore, the detailed study of in vitro digestion and absorption behavior of the EDLAALEK-calcium nanocomposite was performed.结果 Results indicated that the EDLALLEK could bind with Ca2+at a stoichiometric ratio of 1:1. Ca2+coordination induced the self-assembly of the EDLAALEK,resulting in the formation of nanocomposite with crystal structure. Nevertheless,the GI digests of the EDLAALEK-calcium could significantly enhance Ca2+absorption across Caco-2 cell monolayers as compared to the CaCl2.结论 The findings suggest that the EDLAALEK-calcium nanocomposite has the potential as a nutraceutical supplement for improving bone health of the human body.
