目的:观察虎杖总蒽醌对糖尿病肾病(DN)早期血瘀模型大鼠脂代谢及血液流变性的影响。方法:运用链脲佐菌素复制大鼠DN模型。将动物随机分为6组,分别ig高、中、低剂量的虎杖总蒽醌(400,200,100 mg·kg-1),阳性对照组ig开博通6.25 mg·kg-1,模型组和空白组ig同体积(5 mL.只-1)的生理盐水,1次/d,连续8周。实验第8周末,动物取血前1 d,除空白组外,其余各组大鼠sc肾上腺素注射液0.08 mL·kg-1,共2次,间隔4 h,在2次注射间隔期间,将大鼠浸入冰水5 min,以建立DN血瘀模型,并用全自动生化仪、血流变仪分别观察DN早期血瘀模型大鼠脂代谢及血液流变性变化。结果:与模型组比较,虎杖总蒽醌高、中剂量能明显降低TG水平(P<0.01)。高剂量能明显降低LDL水平(P<0.01);与模型组比较,虎杖总蒽醌高、中、低剂量对模型大鼠全血黏度、血浆黏度有明显降低作用。结论:虎杖总蒽醌可降低DN早期血瘀模型大鼠全血黏度和血浆黏度,并对脂代谢紊乱有一定调节作用。
Objective: To observe the effects of total anthraquinones of Polygonum cuspidatum on lipid metabolism and hemorrheology in rats with early stage of blood stasis due to diabetic nephropathy (DN). Methods: The streptozotocin was used to replicate the rat DN model. The animals were randomly divided into 6 groups, high, medium and low doses of Polygonum cuspidatum total anthraquinone (400,200,100 mg · kg-1), the positive control group ig 开通 6.25 mg · kg-1, the model group and the blank group ig The same volume (5 mL. Only -1) of normal saline, 1 / d, for 8 weeks. At the end of the 8th week of experiment, animals were sacrificed 1 d before blood, except for the blank group, the rats in each group were injected with 0.08 mL · kg-1 of sc-epinephrine for 2 times at intervals of 4 h. During the 2 injection intervals, Rats were immersed in ice water for 5 min to establish a model of DN blood stasis. The changes of lipid metabolism and hemorrheology in DN model of early blood stasis were observed by automatic biochemical analyzer and blood rheometer. Results: Compared with the model group, high and medium doses of total anthraquinones of Polygonum cuspidatum could significantly reduce TG levels (P <0.01). Compared with the model group, high, medium and low doses of total anthraquinones of Polygonum cuspidatum significantly reduced the whole blood viscosity and plasma viscosity of model rats. CONCLUSION: Total anthraquinone of Polygonum cuspidatum can reduce whole blood viscosity and plasma viscosity in rats with early blood stasis of DN and play a role in regulation of dyslipidemia.
