为了改进扫描电镜样品制备技术,对比观察了甲醛、戊二醛、戊二醛—锇酸等3种不同固定方法对气管粘膜的固定效果,现简报如下。1 技术过程 按照常规扫描电镜样品的取材和清洗方法,将同一鸡气管的样品切割成适当大小,随机分成3组。第1组用10%甲醛固定;第2组用2.5%戊二醛固定;第3组用戊二醛—锇酸双重固定。固定后,脱水、临界点干燥及喷涂金钯等条件3组均同。用SE—570扫描电镜观察。
