目的:建立测定羟基酪醇含量的高效液相色谱法,考察盐酸水解橄榄苦苷粗提物制备羟基酪醇。方法:采用正交实验,对橄榄苦苷粗提物的水解工艺条件进行考察;以HPLC检测羟基酪醇的含量。结果:最佳水解工艺为:盐酸浓度0.5 mol/L,水解时间15 min,水解温度105℃,料液比1/30,水解后相对于橄榄苦苷的转化率为56.3%,大样经粗提取后羟基酪醇纯度4.6%。结论:该法工艺简单,经济实用,为进一步提高羟基酪醇纯度奠定基础,并为工业化生产羟基酪醇开辟了一条新途径。
OBJECTIVE: To establish a high performance liquid chromatography for the determination of hydroxytyrosol content and study the hydrolysis of oleuropein glycosides from hydrochloric acid to prepare hydroxytyrosol. Methods: The orthogonal experiment was used to study the hydrolysis conditions of crude oleuropein crude extract. The content of hydroxytyrosol was determined by HPLC. Results: The optimum hydrolysis conditions were as follows: hydrochloric acid concentration 0.5 mol / L, hydrolysis time 15 min, hydrolysis temperature 105 ℃, ratio of material to liquid 1/30, hydrolysis rate to oleuropein after hydrolysis was 56.3% After extraction, the purity of hydroxytyrosol was 4.6%. Conclusion: The method is simple, economical and practical, which lays a foundation for further improving the purity of hydroxytyrosol and opens up a new way for the industrial production of hydroxytyrosol.
