目的探讨戊四氮(pentylenetetrazol,PTZ)点燃癫痫大鼠空间学习记忆能力及海马细胞内游离Ca~(2+)浓度([Ca~(2+)]i)、Ca~(2+)/钙调蛋白依赖性蛋白激酶Ⅱα(calcium/calmodulin-dependent protein kinase Ⅱα,CaMKIl_α)mRNA的表达。方法动物分为正常对照组和PTZ组,采用PTZ慢性点燃癫痫模型,应用Morris水迷宫观察2组大鼠空间学习记忆能力,运用流式细胞仪检测海马细胞内[Ca~(2+)]i浓度变化,反转录多聚酶链反应方法测定海马CaMKⅡ_α mRNA的表达。结果 PTZ组大鼠空间学习记忆能力受损,其海马细胞内[Ca~(2+)]i浓度明显升高(P<0.05),CaMKⅡ_αmRNA表达水平明显降低(P<0.05)。结论 PTZ点燃癫痫大鼠海马存在钙超载及CaMKⅡ_α的表达异常,由此引起空间学习记忆受损。
Objective To investigate the effects of pentylenetetrazol (PTZ) on spatial learning and memory ability and intracellular Ca2 + concentration (Ca2 +) i, Ca2 + / Ca2 + The expression of the mRNA of calmodulin-dependent protein kinase Ⅱα (CaMKIl_α). Methods The animals were divided into normal control group and PTZ group. PTZ chronic ignition epilepsy model was used. Morris water maze was used to observe the spatial learning and memory ability of the two groups. Flow cytometry was used to detect the intracellular [Ca ~ (2 +)] i The changes of CaMKⅡ_α mRNA in hippocampus were detected by reverse transcription polymerase chain reaction (RT-PCR). Results The spatial learning and memory abilities of rats in PTZ group were impaired. The concentration of [Ca ~ (2 +)] i in hippocampus was significantly increased (P <0.05) and the expression of CaMKⅡ_α mRNA was significantly decreased in PTZ group (P <0.05). Conclusion There is abnormal calcium overload and CaMKⅡ_α expression in the hippocampus of PTZ-kindled epilepsy rats, which leads to impaired spatial learning and memory.
