利用响应面法对灵芝总三萜的提取工艺进行优化,同时对其抗氧化活性进行研究。在单因素酶解时间、酶解p H、酶解温度和酶用量的基础上,选取纤维素酶、果胶酶和中性蛋白酶的酶用量比例为自变量,灵芝总三萜得率为响应值,进行响应面试验设计,得到最优提取条件为:酶解时间为80 min、提取p H为5、酶解温度50℃、复合酶用量比例为纤维素酶:果胶酶:中性蛋白酶为2.11:0.86:2.09,在该条件下预测值为1.02%、实际值为0.99%、相对误差为2.9%。抗氧化试验中灵芝提取液显示出对DPPH自由基和羟基自由基有一定的清除能力,同时具有较好的还原能力。
