目的探讨第10号染色体缺失的磷酸酶和张力蛋白同源物基因(PTEN)、K-ras蛋白表达与卵巢子宫内膜异位症(内异症)恶变的关系。方法应用免疫组化方法检测21例内异症相关卵巢癌、10例不典型内异症及23例内异症病人PTEN、K-ras蛋白表达,同时结合内异症相关卵巢癌临床病理学参数,分析其在内异症恶变过程中作用。结果内异症相关卵巢癌组PTEN蛋白表达阳性率低于内异症组,差异有显著性(P=0.011);内异症相关卵巢癌组K-ras蛋白阳性表达率高于不典型内异症组及内异症组,差异均有显著性(P=0.002、0.023)。PTEN、K-ras蛋白表达与内异症相关卵巢癌的分化程度、临床分期具有相关性(P=0.007~0.047),而与年龄、病理类型无相关性(P>0.05)。结论 PTEN蛋白表达缺失可能是卵巢内异症恶变过程中的早期事件,而K-ras蛋白的过表达可能与卵巢内异症的恶变有关,PTEN、K-ras的异常表达可能均参与了卵巢内异症的恶变过程。
Objective To investigate the relationship between PTEN, K-ras protein expression and malignant transformation of ovarian endometriosis (endometriosis) on chromosome 10. Methods The expression of PTEN and K-ras protein in 21 cases of endometriosis-related ovarian cancer, 10 cases of atypical endometriosis and 23 cases of endometriosis were detected by immunohistochemistry. Combined with the clinicopathological parameters of endometriosis-associated ovarian cancer , Analysis of its role in endometriosis malignant transformation process. Results The positive rate of PTEN protein expression in endometriosis-related ovarian cancer group was lower than that in endometriosis group (P = 0.011). The positive expression rate of K-ras protein in endometriosis-related ovarian cancer group was higher than that in atypical endometriosis group Syndrome group and endometriosis group, the difference was significant (P = 0.002, 0.023). The expression of PTEN and K-ras was correlated with the differentiation degree and clinical stage of endometriosis-related ovarian cancer (P = 0.007-0.047), but not with age and pathological type (P> 0.05). Conclusions The loss of PTEN protein may be an early event in the malignant transformation of ovarian endometriosis. The overexpression of K-ras protein may be related to the malignant transformation of ovarian endometriosis. The abnormal expressions of PTEN and K-ras may participate in the ovarian Malignant process of the disease.
