黄连超高液相色谱-质谱联用血清药物化学研究

目的建立黄连UPLC-MS/MS血清指纹图谱分析方法,为其质量控制、研究开发和应用提供科学依据。方法采用UPLC-MS/MS技术,通过黄连体外样品、含药血清及空白血清色谱图,确定黄连提取物中大鼠移行成分。结果给药组大鼠血清色谱图与空白色谱图对比有明显不同。含药血清中出现7个移行成分,与黄连提取物色谱图在相同保留时间出峰,为原型成分。采用对照品对照,确定4个色谱峰所表征的化学成分为原型吸收入血成分,依次为黄连碱、表小檗碱、盐酸小檗碱、盐酸巴马汀。通过查阅文献,推断了2个色谱峰所表征的化学成分可能为木兰花碱、格陵兰黄连碱。结论大鼠血中移行成分可能为黄连体内有效物质群,为黄连物质基础研究奠定基础。 OBJECTIVE To establish a method for UPLC-MS / MS fingerprinting of Coptis chinensis to provide a scientific basis for quality control, research, development and application. Methods The UPLC-MS / MS technique was used to determine the components of rats in the extract of Coptidis Rhizoma by HPLC. Results There was a significant difference between the serum chromatogram and the blank chromatogram in the administration group. There were 7 transitional components in the drug-containing serum, and the chromatogram of the extract of Coptis chinensis peaked at the same retention time as the prototype component. Control samples were used to determine the four chromatographic peaks of the chemical composition of the prototype absorption of blood components, followed by berberine, epiberberine, berberine, palmatine hydrochloride. By consulting the literature, we conclude that the chemical constituents of the two peaks may be magnolol and chlorpheniramine. Conclusion The components of blood migration in rats may be effective substances in the body of Coptis chinensis, laying the foundation for the basic research of Coptis.
