目的:观察三七总皂苷片剂对以尿酸钠(MSU)100μg.mL-1诱导血管内皮细胞凋亡的影响。方法:体外培养的血管内皮细胞,以尿酸钠(MSU)100μg.mL-1为凋亡诱导因子,建立血管内皮细胞凋亡诱导模型,继而采用2.5、5、10μg.mL-1浓度的三七总皂苷进行干预,用流式细胞仪测定细胞凋亡率,采用实时定量PCR检测ICAM-1基因的表达。结果:三七总皂苷组2.5μg.mL-1明显抑制细胞凋亡(6.9±1.07)%,与模型组比较P<0.01;能显著抑制ICAM-1基因的表达。结论:三七总皂苷能明显抑制以尿酸钠(MSU)100μg.mL-1诱导的血管内皮细胞凋亡。
Objective: To observe the effects of Panax notoginseng saponins on the apoptosis of vascular endothelial cells induced by sodium urate (MSU) 100μg.mL-1. Methods: Vascular endothelial cells cultured in vitro were induced by 100 ug · mL-1 sodium urate (MSU) as apoptosis-inducing factor, and then induced with 2.5, 5, 10μg.mL-1 The total saponin was intervened, the apoptosis rate was determined by flow cytometry, and the expression of ICAM-1 gene was detected by real-time quantitative PCR. Results: 2.5μg.mL-1 of Panax Notoginseng Saponins could inhibit the cell apoptosis significantly (6.9 ± 1.07%) compared with the model group (P <0.01), and could significantly inhibit the expression of ICAM-1. Conclusion: Panax notoginseng can obviously inhibit the apoptosis of vascular endothelial cells induced by 100 ug · mL-1 sodium urate (MSU).
