目的观察间日疟原虫重组蛋白PvMSP1-19用于检测间日疟患者抗体水平的效果。方法采用His-tag亲和层析柱对间日疟原虫PvMSP1-19重组蛋白初步分离,再经快速蛋白液相色谱仪(FPLC)XK16/70 Superdex75预装柱进一步纯化和Western blot分析。用纯化后的PvMSP1-19重组蛋白包被ELISA板,检测间日疟现症患者、既往感染者和正常人血浆IgG1、IgG4、IgM、IgD、IgE抗体水平。结果间日疟现症患者血浆中抗PvMSP1-19 IgG1抗体水平明显高于间日疟既往感染者和正常人(t=5.535,P<0.05)。结论间日疟现症患者血浆中含有高水平的抗PvMSP1-19抗体IgG1。
Objective To observe the effect of P. vivax recombinant protein PvMSP1-19 on detecting the antibody level of Plasmodium vivax in patients. Methods The recombinant protein of Plasmodium vivax PvMSP1-19 was preliminarily isolated by His-tag affinity chromatography and further purified by fast protein liquid chromatography (FPLC) XK16 / 70 Superdex75 pre-packed column and analyzed by Western blot. The purified PvMSP1-19 recombinant protein was coated on ELISA plates to detect the plasma levels of IgG1, IgG4, IgM, IgD and IgE in patients with P. vivax, formerly infected and normal subjects. The results showed that the level of anti-PvMSP1-19 IgG1 antibody in plasma of patients with P. vivax was significantly higher than that of P. pastivatii and normal controls (t = 5.535, P <0.05). Conclusion The plasma of Plasmodium vivax patients contains high level of anti-PvMSP1-19 IgG1.
