目的检测Wnt5a基因在良性血液病、急性髓细胞白血病及缓解病例骨髓中的表达情况,探讨Wnt5a在急性髓细胞白血病发生中的作用。方法RT-PCR方法检测18例良性血液病病例,30例急性髓细胞白血病初诊病例和25例缓解病例骨髓中Wnt5amRNA的表达情况;免疫细胞化学法检测对应骨髓中Wnt5a蛋白的表达情况。结果Wnt5a mRNA在良性血液病、急性髓细胞白血病病例和缓解病例中的阳性表达率分别为88.9%(16/18)、30%(9/30)和68.0%(17/25),半定量值分别为(0.3826±0.1908)、(0.1041±0.0590)和(0.2606±0.1466)。统计学分析Wnt5a mRNA在初诊病例中的表达明显低于缓解病例(P<0.05),缓解病例与良性血液病之间无明显差异(P>0.05);Wnt5a蛋白在良性血液病中阳性表达,在急性髓细胞白血病初诊病例中低表达或表达缺失,在缓解病例中表达恢复。结论Wnt5a基因在急性髓细胞白血病初诊病例中表达缺失而在治疗缓解病例中表达恢复,提示急性髓系白血病的发生可能与Wnt5a基因表达缺失或下调有关,Wnt5a可以作为一个髓系白血病诊断、指导治疗和疗效评价的指标。
Objective To detect the expression of Wnt5a gene in benign blood diseases, acute myeloid leukemia and remission cases, and to explore the role of Wnt5a in the pathogenesis of acute myeloid leukemia. Methods RT-PCR was used to detect the expression of Wnt5amRNA in 18 cases of benign blood diseases, 30 cases of acute myeloid leukemia and 25 cases of remission cases. The expression of Wnt5a protein in bone marrow was detected by immunocytochemistry. Results The positive rates of Wnt5a mRNA in benign blood diseases, acute myeloid leukemia cases and remission cases were 88.9% (16/18), 30% (9/30) and 68.0% (17/25), respectively. Semiquantitative values (0.3826 ± 0.1908), (0.1041 ± 0.0590) and (0.2606 ± 0.1466), respectively. Statistical analysis Wnt5a mRNA expression in newly diagnosed cases was significantly lower than that in remission cases (P <0.05), there was no significant difference between remission cases and benign blood diseases (P> 0.05); Wnt5a protein was positively expressed in benign blood diseases, Acute myeloid leukemia in newly diagnosed cases of low expression or lack of expression in the ease of expression in patients with recovery. Conclusion Wnt5a gene is expressed in the newly diagnosed cases of acute myeloid leukemia and recovered in the treatment-remitting cases, suggesting that the occurrence of acute myeloid leukemia may be related to the loss or down-regulation of Wnt5a gene expression. Wnt5a can be used as a diagnosis and guide therapy for myeloid leukemia And efficacy evaluation index.
