[目的]观察细胞黏附分子CD146在乳腺癌细胞SKBR3中过表达对裸鼠体内肿瘤生成的影响.[方法]采用脂质体转染乳腺癌细胞株SKBR3,经G418筛选获得过表达CD146阳性克隆细胞,将其接种于裸鼠皮下组织,观察并记录肿瘤生长状况;给肿瘤组织行免疫组织化学染色,观察CD146在肿瘤组织中的表达水平.[结果]将SKBR3细胞经脂质体转染与G418筛选后获得的过表达CD146阳性克隆细胞接种于裸鼠皮下组织后可促进肿瘤组织在裸鼠体内的生成;免疫组织化学染色观察结果表明,接种过表达CD146的SKBR3细胞生成的肿瘤组织仍可高表达CD146蛋白分子.[结论]过表达CD146的乳腺癌细胞SKBR3可促进裸鼠体内的肿瘤生成.
[Objective] To observe the effect of overexpression of cell adhesion molecule CD146 on the tumorigenesis of nude mice in SKBR3 breast cancer cells. [Methods] The liposome-transfected SKBR3 breast cancer cell line was used to screen CD146 positive clones The cells were inoculated into the subcutaneous tissue of nude mice to observe and record the growth of the tumor. Immunohistochemical staining of the tumor tissue was performed to observe the expression level of CD146 in the tumor tissue. [Results] SKBR3 cells were transfected with G418 The overexpression of CD146-positive clonal cells screened after inoculation in nude mice subcutaneous tissue can promote the formation of tumor tissue in nude mice; Immunohistochemical staining results showed that inoculation of SK14 cells over-expression of SKBR3 cells generated tumor tissue can still be high Expressing CD146 protein. [Conclusion] SKBR3, a breast cancer cell overexpressing CD146, can promote tumorigenesis in nude mice.
