本研究旨在研究土槿皮乙酸B(PAB)诱导人乳腺癌MCF-7细胞凋亡和衰老与活性氧的关系。在相差显微镜下观察4μmol/L PAB在36 h诱导细胞凋亡小体的出现;LDH方法显示PAB时间依赖性的增加凋亡比率;SA-β-半乳糖苷酶显示4μmol/L PAB作用3 d后,去药再作用5 d,96%的细胞蓝染,细胞变大且扁平;流式细胞仪检测用DCFH-DA染色后4μmol/L PAB时间依赖性增加细胞内活性氧水平。所以4μmol/L PAB通过增加细胞内活性氧水平促进细胞凋亡和衰老。
The purpose of this study was to investigate the relationship between the apoptosis and senescence of human breast cancer MCF-7 cells induced by hyaluronic acid and acetic acid B (PAB) and reactive oxygen species (ROS). The apoptotic bodies were induced by 4μmol / L PAB at 36 h under phase-contrast microscopy. LDH showed a time-dependent increase in apoptosis rate of PAB. SA-β-galactosidase showed a 4 μmol / L PAB effect for 3 d After 5 days of drug treatment, 96% of the cells were blue-stained, and the cells were enlarged and flattened. The flow cytometry was used to detect the level of reactive oxygen species (ROS) in cells after 4μmol / L PAB treatment with DCFH-DA. Therefore, 4μmol / L PAB promotes apoptosis and senescence by increasing intracellular reactive oxygen species.
