目的研究大鼠皮层神经干细胞定向分化为神经元过程中钙调蛋白(calmodulin,CaM)与肌动蛋白(actin)的时空表达与两者的共存表达。方法采用细胞培养、免疫组化,图像分析术,免疫荧光双标技术检测神经干细胞定向分化为神经元过程中第1、3、5、7天的钙调蛋白与肌动蛋白的时空表达,共焦激光扫描显微镜(CLSM)下观察二者表达共存变化及其相关性。结果在大鼠大脑皮层神经干细胞定向分化的神经元,CaM与Actin的时空表达随着分化时间的增加,表达逐渐增强,随神经元发育逐渐成熟,CaM与Actin的阳性表达也逐渐延伸到突起,分化7天阳性表达最强。CaM的表达与Actin表达基本一致,共存表达胞体和突起。结论在大鼠大脑皮层神经干细胞定向分化的神经元CaM和actin共存表达,随着神经元分化发育成熟表达增强。
Objective To study the spatial and temporal expression of calmodulin (CaM) and actin in rat cortical NSCs differentiated into neurons. Methods The temporal and spatial expressions of calmodulin and actin were detected by cell culture, immunohistochemistry, image analysis and immunofluorescence double labeling on the 1st, 3rd, 5th and 7th day after the neural stem cells were differentiated into neurons Coke laser scanning microscopy (CLSM) under the coexistence of both changes and their correlation. Results The temporal and spatial expression of CaM and Actin in the differentiated neurons of cerebral cortex differentiated gradually increased with the increase of differentiation time. With the maturation of neurons, the positive expression of CaM and Actin gradually extended to the process of neurite outgrowth. The highest positive expression of differentiation 7 days. The expression of CaM was basically the same as that of Actin, which co-existed the expression of soma and neurite. CONCLUSION: CaM and actin are differentially expressed in the differentiated neurons of rat’s cerebral cortex. The expression of CaM and actin is increased with the development of neurons.
