目的 了解TGF β1对增生性瘢痕成纤维细胞中癌基因c myc和c fos的表达及胶原分泌的影响 ,探讨瘢痕增生机制。方法 烧伤患者增生性瘢痕 6例标本 ,通过细胞培养研究了TGF β1对增生性瘢痕成纤维细胞癌基因c myc和c fos的表达 (免疫组织化学和图像分析方法 )及胶原分泌 ( 3 H 脯氨酸法 )的影响。结果 TGF β1可显著提高增生性瘢痕成纤维细胞癌基因c myc和c fos的表达 ,图像分析结果面密度 (c myc :0 0 6 87± 0 0 0 72vs 0 0 14 1± 0 0 0 16 ,P <0 0 1;c fos :0 0 5 2 9± 0 0 0 48vs 0 0 2 2 3± 0 0 0 2 1,P <0 0 1) ;平均灰度 (c myc :3340± 46 2vs 112 0± 2 32 ,P <0 0 1;c fos :14 0 2± 2 86vs 6 0 5± 79,P <0 0 1) ;积分吸光度 [c myc :2 .0± 0 3vs-( 1.5± 0 2 ) ,P <0 0 1;c fos :3.3± 0 4vs -( 1.3± 0 1) ,P <0 0 1]等指标经统计学处理与对照组有显著性差异 ;TGF β1可促使增生性瘢痕成纤维细胞合成分泌胶原率提高 2 0倍 (P <0 0 1)。结论 TGF β1可能是通过癌基因c myc和c fos的表达而促进增生性瘢痕成纤维细胞胶原合成分泌增加。
Objective To investigate the effect of TGFβ1 on the expression of c-myc and c-fos and the secretion of collagen in hypertrophic scar fibroblasts and to explore the mechanism of scar hyperplasia. Methods Six patients with hypertrophic scars were examined for the expression of c-myc and c-fos on the hypertrophic scar fibroblasts (immunohistochemistry and image analysis) and collagen secretion (3 H protamine Acid method) influence. Results TGFβ1 significantly increased the expression of c-myc and c-fos oncogene of hypertrophic scar fibroblasts. The results of image analysis showed that the surface density (c myc: 0 0 6 87 ± 0 0 0 72 vs 0 0 1 14 0 0 0 0 16, P <0 0 1; c fos: 0 0 5 2 9 ± 0 0 0 48 48 0 0 2 2 3 ± 0 0 0 2 1, P <0 0 1); mean gray (c myc: 3340 ± 46 2 vs 112 0 ± 2 32, P <0 0 1; c fos: 14 0 2 ± 2 86 vs 6 0 5 ± 79, P <0 0 1); integral absorbance [c myc: 2.0 ± 0.3vs- (1.5 ± 0 2), P <0 01; c fos: 3.3 ± 0 4vs - (1.3 ± 0 1), P <0 0 1] and other indicators of the treatment group and the control group were significantly different; TGF β1 can promote proliferation Scarring fibroblasts synthesis and secretion of collagen increased 20 times (P <0 0 1). Conclusion TGFβ1 may promote collagen synthesis and secretion of hypertrophic scar fibroblasts through the expression of oncogenes c-myc and c-fos.
