目的体外观察珍珠粉、血竭、乳香、鹿茸、砒石的主要成分(水溶性珍珠粉、血竭素高氯酸盐、乳香水提物、鹿茸多肽、三氧化二砷)对人皮肤成纤维细胞(HSFb)的增殖及胶原蛋白合成的影响,初步探讨其治疗慢性皮肤溃疡的机制。方法组织块法体外培养HSFb;MTT法检测细胞增殖活性;羟脯胺酸比色法测定细胞胶原合成含量。结果药物分别作用于HSFb 48 h后,水溶性珍珠粉在1.25×102~1×103mg/L,血竭素在5.0×100~2.0×101mg/L,乳香水提物在6.4×10-1~2.0×103mg/L,鹿茸多肽在6.25×10-3~4.0×10-1mg/L浓度范围内对HSFb具有促增殖作用,与对照组比较差异显著(P<0.01或P<0.05),三氧化二砷在1.6×10-1~8.0×10-1mg/L浓度范围内对HSFb增殖具有抑制作用,与对照组比较差异显著(P<0.01)。药物分别作用于HSFb 48 h后,水溶性珍珠粉在2.5×102~1×103mg/L,血竭素高氯酸盐在2.5×100~2.0×101mg/L,乳香水提物在6.4×10-1~2.0×103mg/L,鹿茸多肽在1.6×10-3~4.0×10-1mg/L浓度范围内对HSFb胶原蛋白合成具有促进作用,与对照组比较差异显著(P<0.05或P<0.01),三氧化二砷在3.2×10-2~8.0×10-1mg/L浓度范围内对HSFb胶原蛋白合成具有抑制作用,与对照组比较差异显著(P<0.05或P<0.01)。结论敛口生肌药珍珠粉(水溶性珍珠粉)、活血生肌药血竭(血竭素高氯酸盐)、活血生肌药乳香(乳香水提物)、温阳生肌药鹿茸(鹿茸多肽)可促进人皮肤成纤维细胞增殖和胶原合成,蚀疮化腐药砒石(三氧化二砷)抑制人皮肤成纤维细胞增殖和胶原合成。
