目的 采用mRNA差异显示技术 (mRNAdifferentialdisplay,mRNADD) ,寻找MPTP诱导和未诱导的猴中脑黑质组织差异表达的cDNA。 方法 对诱导侧及未诱导侧的黑质mRNA用 3′端 3个锚定引物分别与 5’端 30种随机引物进行逆转录 聚合酶链反应 (RT PCR) ,扩增产物经聚丙烯酰胺凝胶电泳及硝酸银染色显示目的条带。结果 获得一批表达差异的cDNA片段 ,对其克隆后作DNA序列测定、同源性比较分析发现 :其中 2个克隆为未知序列 ,已被GenBank接收 ,接收号为AF15 916 1、AF15 916 2。 结论 MPTP诱导后黑质可能有新基因的表达
Objective To detect cDNAs differentially expressed in the substantia nigra of MPTP-induced and un-induced monkeys by mRNA differential display (mRNADD). Methods RT - PCR was carried out on 3 kinds of random primers in the untreated and untreated side of the substantia nigra by reverse transcription - polymerase chain reaction (RT PCR) using 3 ’3’ anchor primers and 5 ’end respectively. Polymerase chain reaction Gel electrophoresis and silver nitrate staining showed the purpose of the band. Results A number of cDNA fragments were obtained and their DNA sequences were determined after cloning. The homology analysis showed that two of them were unknown and have been accepted by GenBank. The accession numbers are AF15 916 1 and AF15 916 2. Conclusions After MPTP induction, new genes may be expressed in substantia nigra
