目的研究薏米提取物对葡聚糖硫酸钠(DSS)诱导的大鼠溃疡性结肠炎(UC)的预防作用,从抗氧化角度对其作用机制进行初步探究。方法模型组和干预组大鼠自由饮用含5%DSS的水溶液制备UC模型。干预组大鼠用薏米提取物混悬液灌胃,其余两组用等量蒸馏水灌胃。通过结肠大体评分、组织病理学评分、肠壁厚度等评价结肠形态和病理学改变。生化检测大鼠血清和结肠组织髓过氧化物酶(MPO)、超氧化物歧化酶(SOD)和谷胱苷肽过氧化物酶(GSH-Px)活性、丙二醛(MDA)含量及总抗氧化能力(T-AOC)。数据以均数±标准差(x±s)表示,采用SPSS 13.0软件进行单因素方差分析、LSD-t检验,以P<0.05为差异有统计学意义。结果与模型组相比,薏米提取物能明显改善UC大鼠结肠大体形态和组织病理情况,降低血清、结肠组织MPO活性[血清MPO:(103.53±7.36)U/L vs(148.08±16.35)U/L,组织MPO:(2.30±0.34)U/g vs(6.54±2.28)U/g,P均<0.05],能明显降低血清MDA含量[(4.94±0.44)μmol/Lvs(6.83±1.63)μmol/L,P均<0.01]。升高血清和结肠SOD、GSH-Px活性[血清SOD:(225.79±15.86)U/ml vs(200.18±18.42)U/ml,血清GSH-Px:(347.52±20.66)U/ml vs(272.19±7.93)U/ml,P均<0.01];组织SOD:[(2 272.77±485.13)U/ml prot vs(1 116.91±114.41)U/mg prot,组织GSH-Px:(3 094.93±769.62)U/mg prot vs(1 041.64±110.15)U/mg prot,P均<0.05]。结论薏米提取物对DSS诱导的大鼠UC有明显的保护作用,抗氧化是其可能的作用机制之一。
Objective To study the preventive effect of barley extract on dextran sulfate sodium (DSS) -induced ulcerative colitis (UC) in rats and its mechanism of action from the perspective of antioxidation. Methods Rats in model group and intervention group were prepared UC model by freely drinking aqueous solution containing 5% DSS. The rats in the intervention group were given gavage with the suspension of barley extract, and the other two groups were gavaged with the same amount of distilled water. Colonic morphology and pathological changes were evaluated by colon gross score, histopathological score, intestinal wall thickness and the like. The biochemical detection of MPO, SOD and GSH-Px in serum and colon tissues of rats, the contents of malondialdehyde (MDA) and total Antioxidant capacity (T-AOC). Data were expressed as mean ± standard deviation (x ± s), using SPSS 13.0 software for one-way analysis of variance, LSD-t test, P <0.05 was considered statistically significant. Results Compared with the model group, barley extract could significantly improve the colonic morphology and histopathology of UC rats and decrease the activity of MPO in serum and colonic tissue [serum MPO: (103.53 ± 7.36) U / L vs (148.08 ± 16.35) U / (2.30 ± 0.34) U / g vs (6.54 ± 2.28) U / g, all P <0.05], and significantly decreased the level of MDA in serum [(4.94 ± 0.44) μmol / L vs μmol / L, P <0.01]. (SOD: (225.79 ± 15.86) U / ml vs (200.18 ± 18.42) U / ml, serum GSH-Px: (347.52 ± 20.66) U / ml vs (272.19 ± P <0.01). The levels of SOD in the tissue: (2 272.77 ± 485.13) U / ml prot vs (116.91 ± 114.41) U / mg prot and the tissue GSH-Px were (3 094.93 ± 769.62) U / mg prot vs (1041.64 ± 110.15) U / mg prot, P <0.05]. Conclusion The barley extract has a significant protective effect on DSS-induced UC in rats. Antioxidation is one of its possible mechanisms.
