基于上皮细胞-间充质细胞转分化(EMT)理论的艾灸配合化纤Ⅳ号方对实验大鼠TGF-β1干预作用实验研究

目的:基于EMT学说观察EMT-TGF-β1环节化纤Ⅳ号方、艾灸以及二者相配合阻抑实验大鼠肺纤维化的治疗效应,探讨其治疗效应及生物学机制;探讨TGF-β1的变量与EMT病理进程/肺纤维化发展的关联。方法:将鼠龄约为6周的SD大鼠随机分为空白组、模型组、化纤Ⅳ号方组、艾灸组、化纤Ⅳ号方与艾灸配合治疗组(简称为“灸药组”)、化纤Ⅴ号方组,治疗30天后处死观察其肺组织病理改变,并检测其TGF-β1的基因表达情况。结果:实时荧光定量法结果显示:模型组与空白组相比,其TGF-β1m RNA表达显著增高(P<0.001)。与模型组相比,各组的TGF-β1m RNA表达有明显降低(P<0.01)。而各组中,灸药组、化纤Ⅳ号方组疗效最明显,TGF-β1m RNA的表达最低(P<0.001)。但空白组与治疗各组相比,则没有统计学意义。结论:(1)TGF-β1水平的变化与EMT病理进程紧密关联,从而关联到肺纤维化的发展进程;(2)降低TGF-β1对阻逆EMT进程、阻抑肺纤维化发展具有积极意义;(3)化纤Ⅳ号方、艾灸以及二者相配合均在一定程度上具备这种积极作用,其中艾灸配合化纤Ⅳ号方疗法更优;(4)这种治疗效应的生物学机制,可能与通过调控其EMT过程中的TGF-β1m RNA表达关键环节紧密相关。 OBJECTIVE: To observe the therapeutic effect and the biological mechanism of EMT-TGF-β1-link fibrosis Ⅳ, moxibustion and the two in inhibiting pulmonary fibrosis in experimental rats based on the EMT theory, and to explore the therapeutic effect and biological mechanism of TGF-β1 Correlation of variables with EMT pathological progression / pulmonary fibrosis development. Methods: Sprague Dawley rats about 6 weeks old were randomly divided into blank group, model group, chemical fiber IV group, moxibustion group, chemical fiber Ⅳ and moxibustion combined treatment group (abbreviated as “moxibustion group ”), Chemical fiber Ⅴ square group, after treatment for 30 days, sacrificed to observe the pathological changes of lung tissue, and detect the gene expression of TGF-β1. Results: The results of real-time fluorescence quantitative PCR showed that the expression of TGF-β1mRNA in model group was significantly higher than that in blank group (P <0.001). Compared with the model group, the expression of TGF-β1mRNA in each group was significantly decreased (P <0.01). In each group, the curative effect of moxibustion group and chemical fiber group Ⅳ was the most obvious, and the expression of TGF-β1m RNA was the lowest (P <0.001). However, the blank group compared with the treatment group, there is no statistical significance. Conclusions: (1) The change of TGF-β1 level is closely related to the pathological process of EMT, which is related to the development of pulmonary fibrosis; (2) It is of positive significance to decrease the progress of TGF-β1 to obstruct the progression of EMT and inhibit the development of pulmonary fibrosis ; (3) Chemical Fiber Ⅳ, moxibustion and the two are to some extent with this positive effect, of which moxibustion with chemical fiber IV is better; (4) the biological mechanism of this therapeutic effect , Which may be closely related to the key link of TGF-β1m RNA expression through its regulation of EMT.
