目的观察吲哚美辛(IND)对二氧化硅(SiO_2)诱导的肺泡巨噬细胞(AM)中溶菌酶的表达有无抑制作用。方法灌洗大鼠支气管肺泡,将制备好的AM随机分为5组,其中模型组为加入50μg/m L SiO_2粉尘悬液的矽尘中毒模型,干预组在加入SiO_2粉尘悬液的同时分别加入浓度为10-5、10-4和10-3g/m L的IND,对照组给予等体积磷酸盐缓冲液,孵育8 h和16 h后对各组AM进行形态学观察及蛋白免疫印迹实验(WB)。结果干预组AM形态较模型组相对完整,且与IND呈浓度依赖趋势;模型组AM内溶菌酶蛋白表达与对照组相比明显上升(P<0.012 5);孵育8 h后的中、高剂量干预组和孵育16 h后的高剂量干预组AM内溶菌酶蛋白表达面积灰度乘积值与对应模型组相比呈下降趋势(P<0.012 5)。结论 IND可抑制因SiO_2粉尘刺激导致的AM内溶菌酶蛋白表达升高,减轻SiO_2对AM细胞膜的损伤,对AM细胞膜有一定的保护作用。
Objective To observe whether Indomethacin (IND) can inhibit the expression of lysozyme in alveolar macrophages (AM) induced by silicon dioxide (SiO_2). Methods Bronchoalveolar lavage was performed in rats. The prepared AMs were randomly divided into 5 groups. The model group was a silica dust poisoning model with 50 μg / mL SiO 2 dust suspension. The intervention group was added with SiO 2 dust suspension simultaneously IND at the concentrations of 10-5, 10-4 and 10-3g / m L, and the control group was given an equal volume of phosphate buffer. Results The morphology of AM in the intervention group was relatively intact compared with the model group and showed a concentration-dependent trend with IND. Conclusion IND can inhibit the increase of lysozyme protein expression in AM due to the stimulation of SiO_2 dust, reduce the damage of SiO_2 to AM cell membrane, and have certain protective effect on AM cell membrane.
