目的建立一种适合于工业化生产的甘草总黄酮提取和纯化方法。方法以甘草总黄酮的提取率为指标,采用正交试验确定氨水提取的最佳条件;并以甘草总黄酮的转移率和除杂率为指标,采用正交试验优化最佳超滤工艺参数。结果最佳提取条件:0.75%氨水24倍,提取3次,每次60 min,总黄酮平均提取率达83.5%;超滤结果表明:选用10 nm的无机陶瓷膜,在操作压力0.12 MPa和料液温度25℃条件下甘草总黄酮平均转移率和平均除杂率分别达到95.6%和23.3%。结论该联用技术简便可行,生产成本低,适合工业化生产。
Objective To establish a method for the extraction and purification of total flavonoids of licorice suitable for industrialized production. Methods The extraction rate of total flavonoids from licorice root was used as an index to determine the optimum conditions of ammonia extraction by orthogonal test. The orthogonal test was used to optimize the optimum parameters for the ultrafiltration process. Results The optimum extraction conditions were as follows: 0.75% ammonia solution 24 times, extraction 3 times, each time 60 min, the average extraction rate of total flavonoids reached 83.5%; ultrafiltration results show that: the use of 10 nm inorganic ceramic membrane, operating pressure 0.12 MPa and feed The average transfer rate and the average removal rate of total flavonoids of licorice reached 95.6% and 23.3% respectively at the liquid temperature of 25 ℃. Conclusion The combined technology is simple and feasible, low production costs, suitable for industrial production.
