目的探讨女贞苷对Aβ42诱导的神经细胞凋亡及相关蛋白NF-κB、Bcl-2变化的影响及其相关机制。方法人神经母细胞瘤细胞(SH-SY5Y细胞)用女贞苷(50,100μmol·L~(-1))预保护12 h后,加入浓度为15μmol·L~(-1)的Aβ42诱导损伤12 h,用MTT法测定终点细胞存活率,Western blot法检测NF-κB和Bcl-2的蛋白表达变化。结果 50,100μmol·L-1的女贞苷可减少Aβ42引起的SH-SY5Y细胞凋亡。女贞苷组NF-κB的蛋白表达较模型组减少(P<0.01),抗凋亡因子Bcl-2的蛋白表达相对于模型组明显增加(P<0.01)。结论表明女贞苷可提高Aβ42诱导损伤的SH-SY5Y细胞存活率,其机制可能与女贞苷抑制NF-κB的激活和增加抗凋亡因子Bcl-2的蛋白表达有关。
Objective To investigate the effects of virginia glycosides on the apoptosis induced by Aβ42 and the changes of NF-κB and Bcl-2 and related mechanisms. Methods Human neuroblastoma cells (SH-SY5Y cells) were treated with 50 μmol·L -1 pretreatment for 12 h and then induced by Aβ42 at a concentration of 15 μmol·L -1 h. The survival rate of terminal cells was determined by MTT assay. The protein expression of NF-κB and Bcl-2 was detected by Western blot. Results 50, 100μmol·L-1 of virginia glycosides can reduce Aβ42-induced SH-SY5Y cell apoptosis. Compared with the model group, the protein expression of NF-κB decreased in the rosette group (P <0.01), and the protein expression of the anti-apoptotic factor Bcl-2 increased significantly compared with the model group (P <0.01). Conclusions: Ligandrin can enhance the survival rate of SH-SY5Y cells induced by Aβ42, which may be related to the inhibition of the activation of NF-κB and the increase of Bcl-2 protein expression.
