目的:从免疫学角度探讨阿魏菇多糖抑制肿瘤的作用机理。方法:建立昆明小鼠宫颈癌U14移植瘤动物模型,随机分为模型对照组,顺铂作为阳性对照组,阿魏菇多糖组,阿魏菇多糖+顺铂组4组。分别给予不同的药物干预10天后,检测抑瘤率,同时观察阿魏菇多糖对小鼠免疫器官的影响;鸡红细胞半体内法观察阿魏菇多糖对小鼠腹腔巨噬细胞吞噬功能的影响;MTT法测定阿魏菇多糖对小鼠脾脏中NK细胞影响;淋巴细胞转化实验检测阿魏菇多糖对小鼠脾脏T、B淋巴细胞的影响。结果:阿魏菇多糖对小鼠宫颈癌U14移植瘤有抑制作用,抑瘤率达39%,可明显促进荷瘤小鼠腹腔巨噬细胞的吞噬功能,提高脾中NK细胞的免疫活性,增加脾指数,提高脾脏T、B淋巴细胞转化率,与顺铂联用时可提高顺铂对荷瘤鼠免疫功能的抑制。结论:阿魏菇多糖可通过提高小鼠机体免疫功能,抑制小鼠宫颈癌U14移植瘤的增殖。
Objective: To investigate the mechanism of action of polysaccharides from Pleurotus ostreatus inhibiting tumor in immunology. Methods: Kunming mouse cervical cancer U14 xenograft model was established and randomly divided into model control group, cisplatin as positive control group, Pleurotus ferulae polysaccharide group, Pleurotus ferulae polysaccharide + cisplatin group. The effect of polysaccharide from Pleurotus ostreatus on the immune organs of mice was observed after 10 days of different drug intervention. The effect of polysaccharide from Pleurotus eryngii on the phagocytosis of mouse peritoneal macrophages was observed by semi-in vivo method. The effect of polysaccharides from Pleurotus ostreatus on NK cells in spleen of mice was determined by MTT assay. The effect of polysaccharide from Pleurotus ferulae on T and B lymphocytes in spleens of mice was detected by lymphocyte transformation assay. Results: The polysaccharides of Pleurotus ostreatus could inhibit the growth of U14 xenografts in mice, and the tumor inhibition rate was 39%. It could significantly promote the phagocytosis of peritoneal macrophages and increase the immunological activity of NK cells in the spleen Spleen index, improve the T, B lymphocyte transformation rate of the spleen, when combined with cisplatin can improve cisplatin on the immune function of tumor-bearing mice. Conclusion: The polysaccharide of Pleurotus ostreatus can inhibit the proliferation of U14 xenografts in mice by increasing the immune function of mice.
