目的探讨六味安消胶囊及其组分对豚鼠结肠黑变及动力学影响。方法 20只豚鼠随机分为4组:正常组、大黄组、诃子组和六味安消胶囊组,后三组分别给予不同药物灌服30 d。处死动物,检测炭末推进率,观察结肠黑变情况,并对盲肠及近段结肠末染色切片行图像分析。结果大黄组豚鼠结肠有黑变,以盲肠及近段结肠为著。病理改变类似人类。大黄组(87.71%±25.87%)、诃子组(67.63%±19.05%)及六味安消胶囊组(91.14%±16.13%)炭末推进率较正常组(40.38%±6.99%)明显增快(P<0.05)。图像分析显示,诃子组、六味安消胶囊组盲肠、近段结肠固有层色素颗粒灰度及阳性面积比与正常组差异无统计学意义(P>0.05)。大黄组盲肠、近段结肠固有层色素颗粒灰度(盲肠133.18±1.72,结肠137.40±1.39)及阳性面积比(盲肠4.65%±0.76%,结肠2.36%±0.42%)与正常组差异有统计学意义(P<0.01)。结论大黄可致豚鼠实验性结肠黑变病,六味安消胶囊致结肠黑变作用不明显,诃子有预防结肠黑变作用。
Objective To investigate the effects of Liuwei Anxiao capsule and its components on colon blackening and dynamics in guinea pigs. Methods 20 guinea pigs were randomly divided into 4 groups: normal group, rhubarb group, wolfberry fruit group and Liuwei Anxiao capsule group. The latter three groups were given different drugs for 30 days. Animals were sacrificed, charcoal propulsion rate was measured, blackening of the colon was observed, and cecum and near-cold sections of the colon were image-analyzed. [Results] The colon of guinea pigs in the rhubarb group had dark change, with the cecum and the proximal colon. Pathological changes resemble humans. Rhubarb group (87.71%±25.87%), Fructus Gardeniae group (67.63%±19.05%) and Liuwei Anxiao Capsule group (91.14%±16.13%) had a significantly higher rate of charcoal advancement than the normal group (40.38%±6.99%). (P<0.05). Image analysis showed that there was no significant difference in the gray-scale and positive area ratio of pigment particles between the lamina propria and the proximal colon of the jaundice group and Liuwei Anxiao capsule group (P>0.05). In the rhubarb group, there were statistically significant differences in pigment particle gray scales (cecum 133.18±1.72, colon 137.40±1.39) and positive area ratio (cecum 4.65%±0.76%, colon 2.36%±0.42%) between the cecum and the proximal colon, and the normal group. Significance (P<0.01). Conclusion Rhubarb can cause experimental colon melanosis in guinea pigs, and the effect of Liuwei Anxiao Capsule on colon blackening is not obvious. Zongzi can prevent colon blackening.
