目的建立高效分子排阻色谱-蒸发光散射检测法,用于测定右旋糖酐40的分子量及其分布。方法采用TSK-gel G4000PWXL色谱柱(10μm,7.8 mm×300 mm),流动相为水,流速0.6 ml/min,柱温30℃,蒸发光散射检测器。结果右旋糖酐平均重均分子量在4 600~133 800 u范围内线性关系良好(R2=0.991),精密度试验相对标准偏差(RSD)为0.252%,重复性试验RSD为0.460%。结论该方法灵敏度高,重复性好,可作为右旋糖酐40的分子量及其分布测定的分析方法。
OBJECTIVE To establish a high performance liquid chromatography-evaporative light scattering method for the determination of the molecular weight and distribution of dextran 40. Methods TSK-gel G4000PWXL column (10 μm, 7.8 mm × 300 mm) was used. The mobile phase was water and the flow rate was 0.6 ml / min. The column temperature was 30 ℃ and the light scattering detector was used. Results The average weight average molecular weight of dextran was good (R2 = 0.991) in the range of 4 600 ~ 133 800 u, the relative standard deviation (RSD) was 0.252% and the repeatability test RSD was 0.460%. Conclusion The method has high sensitivity and good repeatability and can be used as an analytical method for determining the molecular weight of dextran 40 and its distribution.
