目的:观察络塞维改善D-半乳糖致亚急性衰老大鼠学习记忆力下降的作用,并初步研究其作用机制。方法:将48只SD大鼠随机分为对照组,模型组,阳性药物组,络塞维6、12、24 mg·g-1剂量组。除对照组外,所有大鼠颈部皮下注射D-半乳糖120 mg·kg-1·d-1造模。在给药进行4周后,采用Morris水迷宫测试各试验组大鼠学习记忆能力差异,血气分析仪测定大鼠脑中血氧分压(PO2),血氧浓度(O2)及血氧饱和度(Sa O2),并在试验结束后处死大鼠测定脑组织中超氧化物歧化酶(SOD)、过氧化氢酶(CAT)、谷胱甘肽过氧化酶(GSH-Px)活性和丙二醛(MDA)含量。结果:络塞维12、24 mg·g-1·d-1可显著缩短衰老大鼠在Morris水迷宫的潜伏期(P<0.01或P<0.05),并减少大鼠的游泳总距离和错误角度。同时,给予络塞维6,12,24 mg·g-1·d-1治疗衰老大鼠均能不同程度提高大鼠脑内PO2,O2和Sa O2浓度,SOD、CAT、GSH-Px活性及降低MDA含量,并具一定量效关系。结论:络塞维可改善D-半乳糖所致大鼠学习记忆下降现象,该作用可能与回升血氧含量,并保护SOD、CAT、GSH-Px等酶活性,降低MDA生成有关。
OBJECTIVE: To observe the effect of cloison-wei on improving the learning and memory abilities of sub-acute aging rats induced by D-galactose and to study its mechanism. Methods: Forty-eight Sprague-Dawley rats were randomly divided into control group, model group, positive drug group and nebulized 6,12,24 mg · g-1 dose group. All rats except the control group were injected subcutaneously with D-galactose 120 mg · kg -1 · d -1. Four weeks after administration, Morris water maze test was used to test the learning and memory abilities of rats in each experimental group. Blood gas analyzer was used to measure the partial pressure of oxygen (PO2), oxygen (O2) and oxygen saturation (Sa O2). The rats were sacrificed at the end of the experiment to determine the activities of superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GSH-Px) and malondialdehyde (MDA) content. Results: Cyclodextrin 12, 24 mg · g-1 · d-1 significantly shortened the latent period (P <0.01 or P <0.05) in the Morris water maze and decreased the total swimming distance and wrong angle . At the same time, the rats treated with 4, 12, 24 mg · g-1 · d-1 of nesiritide could improve the levels of PO2, O2 and Sa O2, the activity of SOD, CAT, GSH-Px Reduce MDA content, and with a certain dose-effect relationship. CONCLUSION: Lovibond can improve the decline of learning and memory in rats induced by D-galactose, which may be related to the recovery of blood oxygen content and the activities of SOD, CAT, GSH-Px and MDA.
