目的:建立高效液相色谱法通过示差折光检测器同时测定转化糖电解质注射液中果糖和葡萄糖含量。方法:色谱柱为Waters XbridgeTMAmide(150 mm×4.6 mm,3.5μm),以乙腈-水-三乙胺(75∶25∶0.2)为流动相,柱温为40℃;流速:1.0 ml·min-1;示差折光检测器,检测器温度为40℃,进样量:50μl。结果:果糖在99.8~149.7μg(r=0.999 7)、葡萄糖在100.3~150.4μg(r=0.999 2)范围内与峰面积呈良好的线性关系;果糖和葡萄糖的平均回收率分别为99.2%、99.5%,RSD分别为0.81%、0.84%(n=9)。结论:此法专属性较高,准确度良好且方便快捷,分离效果好,可用于同时测定果糖和葡萄糖的含量。
Objective: To establish a high performance liquid chromatography (HPLC) method for the simultaneous determination of fructose and glucose in invert sugar electrolytes by differential refractometer. METHODS: The column was Waters XbridgeTMAmide (150 mm × 4.6 mm, 3.5 μm) with acetonitrile-water-triethylamine (75:25:0.2) as the mobile phase and the column temperature was 40 ℃. The flow rate was 1.0 ml · min- 1; refractive index detector, detector temperature of 40 ℃, injection volume: 50μl. RESULTS: Fructose had a good linear relationship with peak area in the range of 99.8-149.7 μg (r = 0.999 7) and glucose in the range of 100.3-150.4 μg (r = 0.999 2). The average recoveries of fructose and glucose were 99.2% 99.5% with RSD of 0.81% and 0.84% respectively (n = 9). Conclusion: This method has high specificity, good accuracy and convenience, separation effect is good, can be used for the simultaneous determination of fructose and glucose content.
