目的研究酵母多糖(Zym)致老年全身炎症反应综合征(SIRS)大鼠模型的多器官功能及炎症因子变化。方法壮年和老年SD大鼠,均采用腹腔注射Zym-石蜡混悬液制备SIRS模型。按照不同注射剂量分为750 mg/kg模型组、500 mg/kg模型组、250 mg/kg模型组、125 mg/kg模型组,同时设立空白对照组。观察各组大鼠造模前后的体温、呼吸频率、白细胞计数、心率以确定造模成功。检测空白对照组及各组造模后动脉血气分析指标:pH、二氧化碳分压(PaCO_2)、氧分压(PaO_2)、血氧饱和度(SaO_2),外周血液检测生化指标:丙氨酸氨基转移酶(ALT)、天冬氨酸氨基转移酶(AST)、肌酸激酶(CK)、乳酸脱氢酶(LDH)、肌酐(Cr),以及肿瘤坏死因子-α((TNF-α)、白细胞介素-1β(IL-1β)、白细胞介素-6(IL-6)、白细胞介素-10(IL-10)。计量资料分析采用t检验。结果成年500 mg/kg组和250 mg/kg组血气分析指标、血清学检测指标、炎症因子与空白对照组壮年大鼠比较差异有统计学意义(P<0.01,P<0.05);壮年125 mg/kg组血气分析指标、血清学检测指标、炎症因子与空白对照组壮年大鼠比较,差异无统计学意义(P>0.05)。老年大鼠500 mg/kg组,250 mg/kg组,125 mg/kg组血气分析指标、血清学检测指标、炎症因子与空白对照组老年大鼠比较差异有统计学意义(P<0.01,P<0.01,P<0.05);老年大鼠与壮年大鼠各相同造模剂量组(500 mg/kg组,250 mg/kg组,125 mg/kg组)血气分析指标、血清学检测指标、炎症因子比较差异有统计学意义(P均<0.01)。结论腹腔注射Zym-石蜡混悬液制备壮年大鼠SIRS模型以500 mg/kg为佳,老年大鼠SIRS模型以250 mg/kg为佳。相同剂量Zym老年组各器官功能比壮年组差,炎症因子升高。
Objective To investigate the changes of multiple organs and inflammatory cytokines in the rat model of systemic inflammatory response syndrome (SIRS) induced by zymosan. Methods Male Sprague-Dawley (SD) rats and aged Sprague-Dawley rats were allotted SIRS model with intraperitoneal injection of Zym-paraffin suspension. According to different injection dose, the model group was divided into 750 mg / kg group, 500 mg / kg model group, 250 mg / kg model group and 125 mg / kg model group. At the same time, a blank control group was established. The body temperature, respiration rate, white blood cell count and heart rate of the rats in each group were observed before and after modeling to confirm the success of modeling.
