目的观察T淋巴细胞亚群及连接蛋白43(Cx43)在自发性高血压大鼠(SHR)和WKY大鼠肾脏的表达差异及其与高血压肾损伤的关系。方法 16~17周龄成年雄性SHR和WKY大鼠,各12只,监测尾动脉血压后,分别测定大鼠24h尿蛋白、β_2微球蛋白、24h尿微量白蛋白、血肌酐、尿素氮及尿酸含量,并通过苏木素-伊红(HE)染色进行大鼠肾脏病理学检查,应用流式细胞术检测肾脏中T淋巴细胞亚群以及CD4~+Cx43和CD8~+Cx43的表达比例。结果与WKY大鼠比较,SHR收缩压、24h尿蛋白、β_2微球蛋白、血肌酐、尿素氮和尿酸均明显升高。HE染色结果显示,SHR肾脏血管壁增厚,肾小球出现轻度萎缩,小管上皮细胞出现空泡及颗粒变性,肾间质被炎性细胞浸润等肾损伤现象。与WKY大鼠比较,SHR肾脏中CD4~+和CD8~+T淋巴细胞的比例显著升高,分别为(4.5±0.5)%比(3.2±0.3)%,(4.1±0.3)%比(2.9±0.4)%(均P<0.05);但两种大鼠之间CD4~+/CD8~+的差异无统计学意义(P>0.05)。WKY大鼠和SHR肾脏中CD4~+Cx43占CD4~+T淋巴细胞的比例差异无统计学意义(P>0.05);但SHR肾脏中CD8~+Cx43占CD8~+T淋巴细胞比例高于WKY大鼠[(53.5±5.7)%比(36.5±3.3)%,P<0.05]。结论 SHR肾损伤伴随着肾脏中T淋巴细胞亚群比例上调,且Cx43在CD8~+T淋巴细胞的表达增加。
Objective To investigate the expression differences of T lymphocyte subsets and connexin 43 (Cx43) in the kidney of spontaneously hypertensive rats (SHR) and WKY rats and their relationship with hypertensive renal injury. Methods Twelve adult male SHR and WKY rats aged 16 to 17 weeks old were randomly divided into two groups. After monitoring the tail arterial pressure, the 24h urinary protein, β_2 microglobulin, 24h urinary albumin, serum creatinine, urea nitrogen and uric acid The renal nephrotic pathology was examined by hematoxylin and eosin (HE) staining. The proportion of T lymphocyte subsets and CD4 ~ + Cx43 and CD8 ~ + Cx43 in the kidney were detected by flow cytometry. Results Compared with WKY rats, SHR systolic pressure, 24h urinary protein, β_2 microglobulin, serum creatinine, urea nitrogen and uric acid were significantly increased. The results of HE staining showed that there was thickened blood vessel wall of SHR, mild glomerular atrophy, vacuolization and granular degeneration of renal tubular epithelial cells, renal injury caused by infiltration of inflammatory cells and so on. Compared with WKY rats, the proportion of CD4 ~ + and CD8 ~ + T lymphocytes in SHR kidneys were significantly higher than those in WKY rats (4.5 ± 0.5% vs 3.2 ± 0.3% vs 4.1 ± 0.3% ± 0.4%) (all P <0.05). However, there was no significant difference in CD4 ~ + / CD8 ~ + between the two groups (P> 0.05). The proportion of CD4 ~ + Cx43 in CD4 ~ + T lymphocytes in WKY rats and SHR kidney was not statistically significant (P> 0.05). However, the proportion of CD8 ~ + Cx43 in CD8 ~ + T lymphocytes in SHR kidney was higher than that in WKY Rats [(53.5 ± 5.7)% vs (36.5 ± 3.3)%, P <0.05]. Conclusion The renal injury of SHR is accompanied by an upregulation of the proportion of T lymphocyte subsets in kidney and the increase of Cx43 expression in CD8 + T lymphocytes.
