目的:观察丹参总酚酸对慢性心力衰竭大鼠心肌肌浆网Ca2+-ATP酶(Ca2+-ATPase,SERCA)、受磷蛋白(Phospholamban,PLB)的蛋白表达的影响,探讨丹参总酚酸抗心力衰竭的作用机制。方法:结扎大鼠冠状动脉左前降支造成心肌梗死后慢性心力衰竭模型,连续给予丹参总酚酸4周后,采用蛋白印迹(Western Blot)技术检测心肌组织的SERCA和PLB蛋白表达水平及PLB的磷酸化水平。结果:与心力衰竭模型组相比,丹参总酚酸能明显升高SERCA及磷酸化受磷蛋白(phosphorylated Phospholamban,P-PLB)的蛋白表达及PLB的磷酸化水平。丹参总酚酸能明显降低PLB/SERCA比值。结论:丹参总酚酸能升高SERCA的蛋白表达,升高P-PLB的蛋白表达及PLB的磷酸化水平,降低PLB/SERCA比值,减轻PLB对SERCA功能的抑制,这可能是丹参总酚酸抗心力衰竭的机制之一。
Objective: To observe the effect of total salvianolic acid on the protein expression of Ca2 + -ATPase (SERCA) and phospholamban (PLB) in myocardial sarcoplasmic reticulum of chronic heart failure rats, The mechanism of failure. Methods: The left anterior descending coronary artery of rats was ligated to establish the model of chronic heart failure after myocardial infarction. Continuous salvianolate treatment for 4 weeks followed by Western blotting to detect the expression of SERCA and PLB in myocardium and PLB Phosphorylation levels. Results: Compared with the model group, the total phenolic acids of salvia miltiorrhiza markedly increased the protein expression of SERCA, phosphorylated phospholamban (P-PLB) and the phosphorylation of PLB. Salvia total phenolic can significantly reduce the PLB / SERCA ratio. CONCLUSION: Salvia miltiorrhiza total phenolic acid can increase SERCA protein expression, increase P-PLB protein expression and phosphorylation of PLB, decrease PLB / SERCA ratio, and decrease the inhibitory effect of PLB on SERCA function, One of the mechanisms of heart failure.
