目的 探讨超抗原SEA对Tc1和Tc2亚群细胞增殖的不同影响。方法 以SEA刺激PBMC ,以免疫荧光染色结合流式细胞术比较刺激与未刺激的PBMC中T细胞各亚群比例的变化 ;从PBMC中分离CD8+T细胞 ,定向诱导为Tc1和Tc2细胞系 ,3 H TdR掺入法检测Tc1和Tc2细胞对SEA刺激的反应 ,用免疫荧光染色和流式细胞术检测CD2 5的表达。结果 经SEA刺激后 ,PBMC中的Tc0 /TH0增殖并向Ⅰ型或Ⅱ型分化 ,但SEA所诱导PBMC中的Tc2 /TH2的比例远高于Tc1/TH1;SEA在体外能更有效地促进Tc2亚群细胞的增殖 ;经SEA刺激的Tc2细胞系CD2 5的表达也高于Tc1。结论 SEA优先诱导PBMC中的Tc2 /TH2的分化 ,在体外更有效地促进Tc2细胞增殖
Objective To investigate the different effects of superantigen SEA on the proliferation of Tc1 and Tc2 subpopulations. Methods PBMCs were stimulated with SEA. The proportion of T lymphocytes in stimulated and non-stimulated PBMCs was compared by immunofluorescence staining and flow cytometry. CD8 + T cells were isolated from PBMCs and induced into Tc1 and Tc2 cell lines. The response of Tc1 and Tc2 cells to SEA stimulation was measured by 3 H TdR incorporation assay. The expression of CD25 was detected by immunofluorescence staining and flow cytometry. Results After stimulation with SEA, Tc0 / TH0 in PBMC proliferated and differentiated to type Ⅰ or Ⅱ, but the ratio of Tc2 / TH2 in PBMC induced by SEA was much higher than that in Tc1 / TH1. SEA could promote Tc2 Proliferation of subpopulations; the expression of CD25 on Tc2 cells stimulated by SEA was also higher than that of Tc1. Conclusion SEA preferentially induces the differentiation of Tc2 / TH2 in PBMC and promotes the proliferation of Tc2 cells more effectively in vitro
