目的研究补中益气汤对小鼠肠道上皮细胞天然免疫效应分子β防御素-2(mouseβ-defensin 2,mBD-2)表达的影响。方法取SPF级C57BL/6小鼠,随机分为正常对照组和补中益气汤高、中、低剂量组,给予生理盐水和不同剂量补中益气汤(50,25,12.5 g.kg-1)灌胃,7 d后取小鼠肠道组织,分别用RT-PCR法和免疫组化法检测小鼠肠道上皮细胞mBD-2 mRNA和蛋白表达。结果正常对照组小鼠肠道组织不表达mBD-2。经补中益气汤刺激后,在mRNA水平和蛋白水平均检测到肠道上皮细胞mBD-2的表达,补中益气汤高剂量组作用效果明显,与中、低剂量组比较差异有统计学意义(P<0.05,P<0.01)。结论补中益气汤在体内可以诱导小鼠肠道上皮细胞天然免疫效应分子mBD-2的表达,这可能是其抗炎抑菌的作用机理之一。
Objective To study the effect of Buzhong Yiqi Decoction on the expression of mouse innate immune effector molecule β-defensin-2 (mBD-2) in mouse intestinal epithelial cells. Methods SPF grade C57BL / 6 mice were randomly divided into normal control group and Buzhong Yiqi Decoction high, middle and low dose groups, given saline and different doses of Bu Zhong Yi Qi Tang (50,25,12.5 g.kg -1) were given intragastrically. After 7 days, the intestine tissues of mice were taken out and the expression of mBD-2 mRNA and protein in mouse intestinal epithelial cells was detected by RT-PCR and immunohistochemistry respectively. Results The normal control mice did not express mBD-2 in the gut. After stimulation by Buzhong Yiqi Decoction, the expression of mBD-2 in intestinal epithelial cells was detected at both mRNA and protein levels, and the effect of Buzhong Yiqi Decoction high-dose group was obvious. Compared with the middle-dose and low-dose groups, the difference was statistically significant Significance (P <0.05, P <0.01). Conclusion Buzhong Yiqi Decoction can induce mBD-2 expression in mouse intestinal epithelial cells in vivo, which may be one of the mechanisms of its anti-inflammatory and anti-bacterial effects.
