目的:为诊断戊型肝炎,寻找更好的诊断试剂盒。方法:根据戊型肝炎病毒(HEV)蛋白含有2个可能的抗原表位,从戊型肝炎病毒中国株蛋白的氨基酸序列中选取S30,NS33和S423段抗原肽,用固相合成法进行化学合成,所得目标肽段纯化后经质谱和氨基酸组成分析确证。结果:本文试剂盒与国外品进行比较,血清分析结果表明,前者质量与日本产品相当,明显优于美国Genelabs试剂盒。结论:以这3个合成的抗原肽作为包被抗原组装的戊型肝炎病毒血清抗体的ELISA诊断试剂盒有较好的灵敏性和专一性,可用于临床检测和流行病调查。
Objective: To diagnose hepatitis E, find a better diagnostic kit. Methods: According to the possible epitopes of hepatitis E virus (HEV) protein, S30, NS33 and S423 antigen peptides were selected from the amino acid sequences of the Chinese strain of hepatitis E virus and synthesized by solid-phase synthesis The obtained target peptide was confirmed by mass spectrometry and amino acid composition analysis after purification. Results: The kit was compared with foreign products. The results of serum analysis showed that the quality of the former was similar to that of Japanese products, which was significantly better than that of Genelabs kit from USA. Conclusion: The ELISA diagnostic kit of hepatitis E virus serum with the three synthetic antigen peptides as antigen coating has good sensitivity and specificity and can be used in clinical testing and epidemiological investigation.
