四川省两株脊髓灰质炎Ⅰ型野病毒的核酸序列分析

目的 :探索脊髓灰质炎 (脊灰 )流行与野毒株性状的关系。方法 :选择了 1991年安岳县和 1992年会东县各一例临床诊断为脊灰的患者 ,从其粪便标本中各分离出一株病毒 ,用微量中和试验进行血清学鉴定 ,PCR— RFL P法进行型内鉴定 ,对 VP1区 30 0个核苷酸进行测序。结果 :血清学定型均为脊灰 型病毒 ,经 PCR— RFL P法检测 ,判为野病毒。核酸序列分析证实两株病毒与 Sabin 有明显差异为野病毒。两毒株间核酸序列不完全一致 ,有 2 0个核苷酸的差异。结论 :不同年份、不同地方流行株间的序列有差异。四川省脊灰流行与毒株间核酸序列的差异有关。但我国现行疫苗 OPV能有效预防我省脊灰 ,基础免疫和强化免疫是预防和消灭脊灰的有力武器 Objective: To explore the relationship between polio (polio) prevalence and the characters of wild strains. Methods: One case of poliovirus was selected from one case of Anyue County in 1991 and one case of Huidong County in 1992. One strain of virus was isolated from each stool sample. Serum was identified by micro-neutralization test. PCR-RFL P method for in-type identification, VP1 region 30 nucleotides were sequenced. Results: All the serotypes were polioviruses, which were detected as wild-type virus by PCR-RFLP assay. Nucleotide sequence analysis confirmed that the two viruses and Sabin significant difference for the wild virus. Two strains of nucleic acid sequence is not exactly the same, there are 20 nucleotide differences. Conclusion: There are differences in the sequences between the endemic strains in different years and in different places. The prevalence of polio in Sichuan Province is related to the differences in nucleic acid sequences among strains. However, the current OPV vaccine in our country can effectively prevent polio in our province, and the basic immunity and intensive immunity are powerful weapons to prevent and eradicate polio
