目的:观察中药白疕合剂对体外培养人永生化表皮角质形式细胞(HaCaT细胞)增殖与凋亡的影响,并探讨其作用机制。方法:SD大鼠50只随机分为空白血清组,白疕合剂低、中、高剂量组和阿维A组,用药后制备血清,干预体外培养HaCaT细胞模型,采用MTT法检测白疕合剂对HaCaT细胞增殖的影响;采用流式细胞术检测白疕合剂对HaCaT细胞凋亡的影响。结果:用白疕合剂低、中、高剂量组干预HaCaT细胞,分别培养24 h、48h、72 h后,对HaCaT细胞增殖均有明显抑制作用且呈现浓度依赖性,白疕合剂对HaCaT细胞增殖的抑制作用72h时最强;流式细胞术结果示白疕合剂低、中、高剂量组细胞凋亡率明显增高,凋亡率随药物浓度增高而上升。结论:白疕合剂发挥治疗银屑病的作用,可能与通过抑制HaCaT细胞增殖并诱导其凋亡相关。
Objective: To observe the effect of Baizhen Mixture on the proliferation and apoptosis of human immortalized keratinocytes (HaCaT cells) in vitro and its mechanism. Methods: 50 SD rats were randomly divided into blank serum group, Baijian mixture low, middle and high dose group and Avia A group, after preparation of serum, intervention HaCaT cell model was cultured in vitro, MTT assay was used HaCaT cells proliferation; using flow cytometry Baimai mixture HaCaT cell apoptosis. Results: HaCaT cells were treated with Baizhi Mixture at low, medium and high doses for 24 h, 48 h and 72 h, respectively. HaCaT cells were significantly inhibited and inhibited in a dose-dependent manner. The inhibitory effect was the strongest at 72h. The results of flow cytometry showed that the apoptosis rate of Baijian Mixture group was significantly increased, while the apoptosis rate increased with the increase of drug concentration. Conclusion: The effect of Baizhi Mixture in treating psoriasis may be related to the inhibition of HaCaT cell proliferation and induction of apoptosis.
