通过活细胞计数法、 MTT法、集落形成试验,研究了白头翁水提液(PWE)和醇提液(PAE)的体外抗肿瘤作用。结果:活细胞计数表明 PWE对 7721、Hela、MKN-45细胞株 72 h IC_ (50)分别为 0.88、0.28、0.86 mg/mL,PAE 5 mg/mL对 3种细胞 48 h生长抑制率分别为 78.7%、89.5%、58. 9%;PWE对3种细胞降解MTT IC_(50)值分别为0.82、0.60、0.81mg/mL,PAE 5 mg/mL对 3种细胞降解 MTT抑制率分别为 90. 3%、95. 6%、62.1%;PWE 2 mg/mL对 3种细胞集落抑制率分别为 47. 8%、 63. 4%、 43. 6%; PAE 5 mg/mL对 3种细胞集落抑制率分别为 61. 7%、54.6%、45.3%。提示 PWE、PAE有直接细胞毒作用,并和时间、剂量呈正相关,且 PWE作用比PAE强。
The in vitro anti-tumor effects of P. cinnamomea extract (PWE) and alcohol extract (PAE) were studied by means of live cell counting, MTT assay and colony formation assay. RESULTS: The viable cell counts showed that the IC50 of PWE to 7721, HeLa, and MKN-45 cell lines was 0.88, 0.28, and 0.86 mg/mL respectively for 72 h, and PAE 5 mg/mL for 3 cells. h The growth inhibition rates were 78.7%, 89.5%, and 58. 9%; MTT IC_(50) values of PWE on three cell types were 0.82, 0.60, and 0.81 mg/mL, respectively, and PAE 5 mg/mL had 30% MTT inhibition rate on three cell types. 3%, 95. 6%, 62.1%; PWE 2 mg/mL inhibited the colony of the three kinds of cells were 47. 8%, 63. 4%, 43. 6%; PAE 5 mg/mL inhibited the colony of the three kinds of cells were 61. 7%, 54.6%, 45.3%. It is suggested that PWE and PAE have direct cytotoxicity and are positively correlated with time and dose, and PWE is more potent than PAE.
