目的观察卵泡刺激素(FSH)对卵巢癌细胞株 SKOV-3和 ES-2细胞增殖、凋亡和迁移侵袭的影响。方法将 FSH 以不同浓度(10、20、40、80、160mU/ml)分别作用于1×10~5或2.5×10~6个 SKOV-3细胞和6×10~4或1.5×10~6个 ES-2细胞,FSH 0mU/ml 为对照组,其中 FSH 作用于 SKOV-3细胞的适宜时间为48h,ES-2细胞为24h,分别测定细胞的增殖活性;细胞凋亡和细胞周期情况;细胞培养上清液中基质金属蛋白酶2(MMP-2)的活性;SKOV-3和 ES-2细胞内 MMP-2蛋白及 mRNA 表达的变化;细胞的迁移侵袭能力。结果 (1)随着 FSH 浓度的增加,SKOV-3细胞的增殖活性升高,与对照组相比差异均有统计学意义(均 P<0.01)。(2)随着 FSH 浓度的增加,SKOV-3细胞的凋亡率下降,分别为0.94%±0.06%、0.71%±0.03%、0.22%±0.02%、0.32%±0.02%、0.55%±0.05%,与对照组(1.30%±0.10%)比,差异均有统计学意义(均 P<0.01)。(3)FSH 浓度自40mU/ml 起,SKOV-3细胞 G_0/G_1期百分率逐渐下降,S 期百分率逐渐上升,与对照组比差异均有统计学意义(均P<0.05)。(4)FSH 可提高 SKOV-3细胞 MMP-2 mRNA 和胞质中 MMP-2蛋白含量及细胞培养上清液中 MMP-2的恬性。(5)侵袭实验:SKOV-3细胞加 FSH 组和对照组穿透基底膜细胞数分别为(157±20)、(27±9)个/高倍镜(×200),两组比较差异有统计学意义(P<0.01);划痕实验也显示 FSH 提高了 SKOV-3细胞的迁移能力。FSH 作用于 ES-2细胞的表现与 SKOV-3细胞基本一致。结论 FSH 可促进卵巢癌细胞株 SKOV-3和 ES-2细胞增殖、迁移和侵袭,抑制细胞的凋亡。
Objective To observe the effects of follicle stimulating hormone (FSH) on the proliferation, apoptosis and migration of ovarian cancer cell lines SKOV-3 and ES-2. Methods FSH was administered to 1 × 10 ~ 5 or 2.5 × 10 ~ 6 SKOV-3 cells and 6 × 10 ~ 4 or 1.5 × 10 ~ 6 cells in different concentrations (10,20,40,80,160mU / ml) ES-2 cells and FSH 0mU / ml were control group. (5) Invasion experiment: The number of cells penetrating the basement membrane in SKOV-3 cells plus FSH group and control group were (157 ± 20) and (27 ± 9) / high magnification lens (× 200) respectively. The performance of FSH on ES-2 cells was basically the same as that of SKOV-3 cells. Conclusion FSH can promote the proliferation, migration and invasion of ovarian cancer cell line SKOV-3 and ES-2 and inhibit the apoptosis of ovarian cancer cell line SKOV-3 and ES-2.
