目的探讨紫杉醇对人骨肉瘤细胞株MG-63的增殖抑制及诱导凋亡作用。方法用不同浓度的紫杉醇(0.1、0.5和1μmol/L)作用于人骨肉瘤MG-63细胞12小时、24小时和48小时后,MTT方法检测紫杉醇对MG-63细胞的增殖抑制效果;流式细胞仪检测细胞凋亡;Caspase活性定量检测试剂盒检测半胱天冬特异性蛋白酶-9,3(Caspase-9,3)的活性;探讨紫杉醇对体外培养的人骨肉瘤细胞增殖、凋亡的影响与可能机制。结果紫杉醇对MG-63细胞增殖具有抑制作用,其增殖抑制作用随作用时间延长及药物浓度升高更加明显,具有时间和剂量依赖性;流式细胞仪检测可见紫杉醇能诱导细胞发生凋亡;Caspase活性定量检测MG-63细胞内Caspase-9,3的活性增加。结论紫杉醇可以通过时间依赖性和剂量依赖性方式,抑制M G-63增殖,诱导其凋亡。这种凋亡可能是Caspase-3依赖性的。
Objective To investigate the inhibitory effect of paclitaxel on the proliferation of human osteosarcoma cell line MG-63 and the induction of apoptosis. Methods Paclitaxel (0.1, 0.5 and 1 μmol / L) was used to treat human osteosarcoma MG-63 cells for 12 hours, 24 hours and 48 hours. MTT assay was used to detect the inhibitory effect of paclitaxel on MG-63 cells. Flow cytometry The activity of Caspase-9, 3 was detected by Caspase activity assay kit. The effect of paclitaxel on the proliferation and apoptosis of human osteosarcoma cells in vitro was analyzed. Possible mechanism. Results Paclitaxel had an inhibitory effect on the proliferation of MG-63 cells. The inhibition of proliferation was more pronounced with time and dose-dependently with the prolongation of action time and concentration of paclitaxel. Paclitaxel induced the apoptosis of cells by flow cytometry. Caspase The activity of Caspase-9,3 in MG-63 cells was increased by the activity assay. Conclusion paclitaxel can inhibit the proliferation of M G-63 and induce its apoptosis in a time-dependent and dose-dependent manner. This apoptosis may be Caspase-3 dependent.
