[目的]观察NF-κB抑制剂吡咯烷二硫代氨基甲酸(pyrrolidine dithiocarbamate,PDTC)对顺铂毒性的影响。[方法]SD大鼠40只,随机分为4组,对照组和PDTC组各10只连续用药13 d,于第14天处死;顺铂组和PDTC-顺铂联合用药组各10只连续给药5 d后处死;测定体重和肾重、血肌酐、血尿素氮和白细胞数,用放射免疫法检测血清及肾组织匀浆TNF-α、IL-1含量。[结果]顺铂组大鼠肾系数增大为4.9440±0.5456,BUN升高至(61.073±44.782)mg/dL,血Cr增高至(9.743±3.821)mg/dL,肾组织匀浆IL-1含量升高;顺铂合用PDTC组的大鼠肾系数为4.4517±0.3658,BUN为(59.575±27.914)mg/dL,血Cr为(8.322±2.855)mg/dL,肾组织匀浆IL-1含量近正常。两组间肾IL-1含量比较,差异有显著性意义(P<0.05)。[结论]PDTC不能明显降低顺铂引起的肾毒性。
[Objective] To investigate the effect of pyrrolidine dithiocarbamate (PDTC), an inhibitor of NF-κB, on cisplatin toxicity. [Methods] Forty Sprague-Dawley rats were randomly divided into 4 groups. The control group and PDTC group were given 10 consecutive days for 13 days, and were sacrificed on the 14th day. The rats in the cisplatin group and the PDTC- The rats were killed after 5 days of treatment. The body weight, kidney weight, serum creatinine, blood urea nitrogen and white blood cells were measured. The content of TNF-α and IL-1 in serum and kidney homogenate were measured by radioimmunoassay. [Results] The renal coefficient of cisplatin group was 4.9440 ± 0.5456, the BUN increased to (61.073 ± 44.782) mg / dL, the blood Cr increased to (9.743 ± 3.821) mg / dL, the renal homogenate IL- 1 (P <0.05). The renal coefficient of cisplatin combined with PDTC group was 4.4517 ± 0.3658, the BUN was (59.575 ± 27.914) mg / dL and the blood Cr was (8.322 ± 2.855) mg / dL Near normal. The level of renal IL-1 between the two groups, the difference was significant (P <0.05). [Conclusion] PDTC can not significantly reduce cisplatin-induced nephrotoxicity.
