研究旨在探讨显微注射剂量对鸡胚孵化和转基因效率的影响。将SB转座子介导的转基因载体分别设置为20 ng、50 ng和100 ng,通过显微注射技术注射至14HH鸡胚血管,继续孵化至出雏。检测孵化第7、14和21天的鸡胚成活率,出雏率、初生体重和性腺阳性率。结果表明,50 ng剂量组鸡胚性腺阳性率明显高于其它两组,且各剂量组间绿色荧光蛋白基因表达强度差异不显著。各剂量组之间种蛋成活率、出雏率和雏鸡初生重差异均不显著(P>0.05)。本研究为鸡胚显微注射剂量优化提供参考,对提高转基因鸡制备效率具有重要意义。
The aim of this study was to investigate the effects of microinjection dosages on hatch and transgene efficiency in chicken embryos. The SB transposon-mediated transgene vector was set at 20 ng, 50 ng and 100 ng, respectively, and injected into 14HH chick embryo vessels by microinjection to hatch until hatching. The survival rate, hatching rate, birth weight and gonadal positive rate on the 7th, 14th and 21th day of hatching were detected. The results showed that the positive rate of gonads in 50 ng dose group was significantly higher than that in the other two groups, and there was no significant difference in the expression intensity of GFP between each dose group. There was no significant difference in egg survival rate, hatching rate and chick newborn weight between each dose group (P> 0.05). This study provides a reference for optimizing the dose of chicken embryo microinjection, which is of great significance to improve the efficiency of preparation of transgenic chicken.
