目的 探讨二甲双胍对实验性脂肪肝的治疗作用。 方法 通过12周高脂饲料喂养制备大鼠实验性脂肪肝模型;肝脏病理切片证实造模成功后,取24只造模大鼠随机分为3组并给予相应处理:二甲双胍组给予二甲双胍156mg/kg灌胃,1次/d;饮食治疗组及模型对照组等容积蒸馏水灌胃;除模型对照组外,其余各组均进食普通饲料。4周后分别检测各组动物的肝功能、血脂、血糖、肝脂质、肝指数胰岛素抵抗指数及肝组织学改变。 结果 二甲双胍能显著降低血脂、肝脂,改善肝功能;而饮食治疗使肝组织学有一定改善,但肝酶活性、血清甘油三酯和胆固醇增高持续存在。 结论 二甲双胍可有效地用于实验性脂肪肝的治疗。
Objective To investigate the therapeutic effect of metformin on experimental fatty liver. Methods The rat model of fatty liver was prepared by feeding for 12 weeks with high fat diet. After the pathological section of liver was confirmed to be successful, 24 model rats were randomly divided into 3 groups and given corresponding treatment: metformin group was given metformin 156mg / kg Gavage, 1 / d; diet treatment group and model control group, the same volume of distilled water; in addition to the model control group, the remaining groups were fed ordinary feed. After 4 weeks, liver function, blood lipid, blood glucose, liver lipid, index of liver index and liver histology were detected in each group. Results Metformin can significantly reduce blood lipids, liver fat, improve liver function; and diet therapy to improve liver histology, but liver enzyme activity, serum triglyceride and cholesterol persist. Conclusion Metformin can be effectively used in the treatment of experimental fatty liver.
