目的 探讨豚鼠庆大霉素耳毒性与肾毒性的关系。方法 通过ABR测试 ,耳蜗铺片毛细胞计数 ,血液庆大霉素药代动力学分析 ,血BUN、Cr值测定 ,肾标本光镜下观察等方法 ,观察肌注庆大霉素后豚鼠的耳蜗功能及肾功能变化。结果 肌注庆大霉素二周组ABR的IV波反应阈阈移明显高于肌注庆大霉素一周组及其生理盐水对照组 (前者P <0 .0 5 ,后者P <0 .0 1)。光镜下耳蜗铺片毛细胞计数二周组毛细胞缺失数明显多于一周组和对照组。肌注庆大霉素二周组的血清庆大霉素清除率明显低于生理盐水对照组和肌注庆大霉素一周组 (P <0 .0 5 )。各组实验动物的血清BUN和Cr水平均无明显差异 (P >0 .0 5 ) ,光镜下各组动物的肾小球、近曲小管结构也无明显异常。结论 庆大霉素耳毒性的发生早于肾毒性 ,两者之间不存在相关性
Objective To investigate the relationship between gentamicin ototoxicity and nephrotoxicity in guinea pigs. Methods The cochlea of guinea pigs after intramuscular injection of gentamicin were observed by ABR test, hair cell count of cochlear implants, pharmacokinetics of gentamicin in blood, determination of blood BUN and Cr, and observation of renal specimens by light microscope. Function and renal function changes. Results The IV wave threshold threshold of ABR was significantly higher than that of intramuscular injection of gentamicin for one week and saline control group (P <0.05 for the former and P <0 for the latter). 0 1). Conclusion Gentamicin ototoxicity occurs earlier than nephrotoxicity, and there is no correlation between them
