黄芪对糖尿病大鼠肾皮质TGF-β表达的影响

目的观察黄芪对糖尿病肾病(DN)的作用。方法以STZ诱导糖尿病动物模型,采用免疫组化和RTPCR检测方法观察黄芪对糖尿病大鼠肾皮质TGFβ蛋白、基因表达的影响。结果糖尿病大鼠肾皮质TGFβ免疫染色强度比未治疗大鼠明显减轻,TGFβmRNA水平比未治疗大鼠明显下降(P<0.01),黄芪与开搏通合用可增强上述作用。结论黄芪可抑制糖尿病大鼠肾皮质TGFβ的过度表达,其防治DN发生、发展的作用机制可能部分是通过抑制TGFβ的过度表达。 Objective To observe the effect of Astragalus on diabetic nephropathy (DN). Methods STZ-induced diabetic animal models were used to observe the effect of astragalus on renal cortex TGFβprotein and gene expression in diabetic rats by immunohistochemistry and RTPCR. Results The intensity of TGFβ immunoreactivity in renal cortex of diabetic rats was significantly lower than that of untreated rats. The level of TGFβ mRNA was significantly lower than that of untreated rats (P<0.01). The combination of jaundice and captopril enhanced the above effects. Conclusion Astragalus can inhibit the overexpression of TGF-β in the renal cortex of diabetic rats. The mechanism of its action may be partly through inhibiting the overexpression of TGFβ.
