目的 克隆 1、2、3、4周龄小鼠睾丸之间差异表达的基因。 方法 应用改良的mRNA差异显示技术 ,对 1、2、3、4周龄小鼠睾丸进行了基因表达的差异显示分析 ,并对其中 1个从 2周龄小鼠中高表达的cDNA片段进行了克隆和测序。 结果 所克隆的cDNA片段与小鼠大脑、造血干细胞p38MAPK(p38betamitogen activatedpro teinkinase ,p38MAPK β)基因的同源性分别为 91%和 85 %。Northerndotblot结果显示该基因在小鼠睾丸和脑组织中表达最高。 结论 小鼠p38MAPK基因可能与小鼠精子发生相关
Objective To clone the genes differentially expressed in testis of 1, 2, 3 and 4 weeks old mice. Methods The differential display of gene expression in testis of 1, 2, 3, 4 weeks old mice was demonstrated by the improved mRNA differential display technique. One of them was cloned from the cDNA of 2 weeks old mouse which was overexpressed And sequencing. Results The cloned cDNA fragment showed 91% and 85% homology with the p38MAPK (p38MAPKβ) gene of mouse brain and hematopoietic stem cells, respectively. The results of Northerndotblot showed that the gene was most expressed in the testis and brain of mice. Conclusion Mouse p38MAPK gene may be related to mouse spermatogenesis
