目的研究肿瘤浸润转移相关基因粘附分子吞噬糖蛋白(CD44v6)、基质金属蛋白酶组织抑制物-1(TIMP-1)在子宫颈鳞状细胞癌中的表达及意义。方法应用免疫组化S-P法检测子宫颈癌手术切除组织60例,正常组织10例,观察肿瘤转移相关基因CD44、TIMP-1的表达与子宫颈鳞状细胞癌发生及浸润、转移的关系。结果(1)60例子宫颈鳞状细胞癌组织中,CD44、TIMP-1的表达阳性检出率分别为:51.7%(31/60)、31.7%(19/60)。(2)CD44在不同组织学分级和有、无淋巴结转移的子宫颈鳞状细胞癌患者中表达差异有显著性,P<0.05。TIMP-1在不同浸润深度子宫颈鳞状细胞癌组织中表达差异有显著性,P<0.05,TIMP-1在晚期高表达,提示可作为联合检测肿瘤负荷的标志物之一。结论提示CD44蛋白的低表达与子宫颈癌的病情发展及转移的发生相关,并提示可作为预后判定的参数。TIMP-1的失活对子宫颈鳞状细胞癌获得浸润转移的潜能可能起着重要作用,并提示可作为肿瘤的分期指标。
Objective To study the expression and significance of CD44v6 and TIMP-1 in cervical squamous cell carcinoma of the cervix. Methods Sixty cases of cervical cancer and 10 cases of normal tissue were examined by immunohistochemical S-P method. The relationship between the expression of CD44, TIMP-1 and the occurrence and infiltration of cervical squamous cell carcinoma was observed. Results (1) The positive rates of CD44 and TIMP-1 expression in 60 cases of cervical squamous cell carcinoma were 51.7% (31/60) and 31.7% (19/60), respectively. (2) The expression of CD44 was significantly different in patients with cervical squamous cell carcinoma with different histological grading and without lymph node metastasis (P <0.05). The expression of TIMP-1 in different invasive depth of cervical squamous cell carcinoma tissue was significantly different, P <0.05, TIMP-1 highly expressed in the late stage, suggesting that as a combined detection of tumor burden one of the markers. The results suggest that the low expression of CD44 protein is associated with the progression of cervical cancer and metastasis, and may be used as a prognostic parameter. The inactivation of TIMP-1 may play an important role in the invasion and metastasis of squamous cell carcinoma of the cervix, and may be used as a staging indicator of tumor.
