目的 比较SARS冠状病毒各分离株S基因序列及氨基酸序列之间的差异 ,预测SARS -CoV的S蛋白的抗原表位。方法 利用Lasergene软件包中的EditSeq从 2 1株SARS冠状病毒分离株截取S基因序列并翻译成氨基酸序列 ,然后用ClustalX软件对截取序列进行比较分析 ,确定基准株 ,最后用Protean软件对基准株进行抗原表位预测。结果 在 2 1株分离株的S基因中有 8株核苷酸序列、13株氨基酸序列没有发生点突变 ;预测出 78个可能性抗原表位。结论 SARS -CoV的S蛋白相当保守 ,只发生个别点突变 ,可能性抗原表位多 ,且在其中的 14个区域中可能性最显著。
Objective To compare the differences in S gene sequences and amino acid sequences between SARS coronavirus isolates and to predict the epitope of S protein of SARS-CoV. Methods The sequence of S gene was cloned from 21 SARS coronavirus isolates using EditSeq in Lasergene software package and translated into amino acid sequence. The sequence was compared with ClustalX software to determine the reference strain. Finally, the reference strain was performed with Protean software Epitope prediction. Results There were 8 nucleotide sequences in S gene of 21 isolates and no point mutation in 13 amino acid sequences. 78 potential epitopes were predicted. Conclusion The S protein of SARS-CoV is rather conservative with only some point mutations. The possibility of epitopes is very high, and the most likely of them is in 14 of them.
