目的:探讨自噬基因beclin-1在儿童急性淋巴细胞白血病(ALL)患者外周血白细胞的表达及意义。方法:选取儿童急性淋巴细胞白血病患者39例,体检正常儿童40例作为对照,收集患者及正常对照外周血标本,分离白细胞,用免疫组化和免疫印迹法检测beclin-1表达,分析比较患者与对照组beclin-1蛋白表达差异。结果:ALL患儿外周血白细胞beclin-1表达阳性率48.7%(19/39),低于正常对照组82.5%(33/40)(P<0.005),Western blot显示患儿阳性标本表达量亦低于对照阳性标本。标危组表达率(59.3%)高于高危组(25.0%),B-ALL与T-ALL的beclin-1表达率差异无显著性(P<0.05)。结论:beclin-1表达率及表达量在ALL患儿外周血白细胞中明显低于正常对照;beclin-1表达率与诊断时免疫学分型无明显关系,标危组表达率高于高危组。beclin-1可能与儿童ALL发生发展相关。
Objective: To investigate the expression and significance of autophagy gene beclin-1 in peripheral leukocytes in children with acute lymphoblastic leukemia (ALL). Methods: Thirty-nine children with acute lymphoblastic leukemia and 40 normal children were enrolled in this study. Peripheral blood samples were collected from patients and normal controls. Leukocytes were isolated and the expression of beclin-1 was detected by immunohistochemistry and Western blotting. Control group beclin-1 protein expression differences. Results: The positive rate of beclin-1 in ALL children was 48.7% (19/39), which was lower than that in normal controls (82.5%, 33/40) (P <0.005). Western blot showed that positive expression of beclin-1 Lower than the control positive specimens. The expression rate of beclin-1 in B-ALL group and T-ALL group was significantly higher than that in high-risk group (59.3% vs 25.0%, P <0.05). CONCLUSION: The expression and expression of beclin-1 in peripheral leukocytes of children with ALL was significantly lower than that of normal controls. The expression of beclin-1 was not related to the immunological classification at diagnosis. The expression rate of beclin-1 was higher than that of high-risk group. Beclin-1 may be associated with the development of childhood ALL.
